NM_002485.5(NBN):c.2138C>G (p.Ala713Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A713G variant (also known as c.2138C>G), located in coding exon 14 of the NBN gene, results from a C to G substitution at nucleotide position 2138. The alanine at codon 713 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
