Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.5503G>A (p.Glu1835Lys), citing Ambry Variant Classification Scheme 2023: The p.E1835K variant (also known as c.5503G>A), located in coding exon 43 of the CACNA1C gene, results from a G to A substitution at nucleotide position 5503. The glutamic acid at codon 1835 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
