Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_201596.3(CACNB2):c.544A>G (p.Met182Val), citing Ambry Variant Classification Scheme 2023: The p.M128V variant (also known as c.382A>G), located in coding exon 4 of the CACNB2 gene, results from an A to G substitution at nucleotide position 382. The methionine at codon 128 is replaced by valine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
