NM_001134363.3(RBM20):c.434G>A (p.Gly145Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G145D variant (also known as c.434G>A), located in coding exon 2 of the RBM20 gene, results from a G to A substitution at nucleotide position 434. The glycine at codon 145 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
