This variant lies in the APC gene (transcript NM_000038.6) at coding-DNA position 8353, where A is replaced by C; at the protein level this means replaces asparagine at residue 2785 with histidine — a missense variant. Submitter rationale: The APC c.8353A>C (p.N2785H) variant has been reported in at least one individual with breast cancer (PMID: 30093976 ). It was observed in 6/19910 chromosomes of the East Asian subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 411435). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
