NM_000038.6(APC):c.2411A>G (p.Asn804Ser) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): This variant is denoted APC c.2411A>G at the cDNA level, p.Asn804Ser (N804S) at the protein level, and results in the change of an Asparagine to a Serine (AAT>AGT). This variant has been observed in at least one individual with breast cancer (Tung 2015). APC Asn804Ser was not observed at a significant allele frequency in large population cohorts (Lek 2016). This variant is not located in a known functional domain. In silico analyses predict that this variant is unlikely to alter protein structure or function. Based on currently available evidence, it is unclear whether APC Asn804Ser is a pathogenic or benign variant. We consider it to be a variant of uncertain significance.
