Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.52A>G (p.Ser18Gly), citing Ambry Variant Classification Scheme 2023: The p.S32G variant (also known as c.94A>G), located in coding exon 2 of the MUTYH gene, results from an A to G substitution at nucleotide position 94. The serine at codon 32 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
