NM_001048174.2(MUTYH):c.844C>G (p.Gln282Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q310E variant (also known as c.928C>G), located in coding exon 10 of the MUTYH gene, results from a C to G substitution at nucleotide position 928. The glutamine at codon 310 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
