Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.778A>T (p.Thr260Ser), citing Ambry Variant Classification Scheme 2023: The p.T288S variant (also known as c.862A>T), located in coding exon 10 of the MUTYH gene, results from an A to T substitution at nucleotide position 862. The threonine at codon 288 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
