NM_001048174.2(MUTYH):c.1143G>C (p.Trp381Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W409C variant (also known as c.1227G>C), located in coding exon 13 of the MUTYH gene, results from a G to C substitution at nucleotide position 1227. The tryptophan at codon 409 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
