Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.1053G>T (p.Gln351His), citing Ambry Variant Classification Scheme 2023: The p.Q379H variant (also known as c.1137G>T), located in coding exon 12 of the MUTYH gene, results from a G to T substitution at nucleotide position 1137. The glutamine at codon 379 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
