Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.61G>T (p.Gly21Trp), citing Ambry Variant Classification Scheme 2023: The p.G35W variant (also known as c.103G>T), located in coding exon 2 of the MUTYH gene, results from a G to T substitution at nucleotide position 103. The glycine at codon 35 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
