Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.917C>A (p.Pro306His), citing Ambry Variant Classification Scheme 2023: The p.P334H variant (also known as c.1001C>A), located in coding exon 12 of the MUTYH gene, results from a C to A substitution at nucleotide position 1001. The proline at codon 334 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
