Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001927.4(DES):c.700G>A (p.Glu234Lys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: DES c.700G>A (p.Glu234Lys) results in a conservative amino acid change located in the Intermediate filament, rod domain (IPR039008) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251444 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.700G>A has been reported in the literature in one individual affected with Cardiomyopathy, however this individual also carried variants of uncertain significance in other genes (MYPN and CACNA1C) that may contribute to the observed phenotype (Chen_2017). Therefore, this report does not provide unequivocal conclusions about association of the variant with Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One ClinVar submitter has assessed the variant since 2014: the variant was classified as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
