NM_000179.3(MSH6):c.2308G>C (p.Gly770Arg) was classified as Likely pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G770R variant (also known as c.2308G>C), located in coding exon 4 of the MSH6 gene, results from a G to C substitution at nucleotide position 2308. The glycine at codon 770 is replaced by arginine, an amino acid with dissimilar properties. This variant has been identified in a proband whose Lynch syndrome-associated tumor demonstrated loss of MSH6 expression by immunohistochemistry (Li S et al. J. Med. Genet. 2020 Jan;57:62-69; Ambry internal data).This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
