NM_006030.4(CACNA2D2):c.485_486del (p.Tyr161_Tyr162insTer) was classified as Pathogenic for Early-infantile DEE by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change deletes 2 nucleotides in exon 5 of the CACNA2D2 mRNA (c.485_486delAT), causing a frameshift at codon 162. This creates a premature translational stop signal (p.Tyr162*) and is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. While this particular variant has not been reported in the literature, truncating variants in CACNA2D2 are known to be pathogenic (PMID: 24358150).
