Pathogenic for Retinoblastoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000321.3(RB1):c.1673_1674dup (p.Glu559fs), citing Invitae Variant Classification Sherloc (09022015): While this particular variant has not been reported in the literature, truncating variants in RB1 are known to be pathogenic (PMID: 17096365). For these reasons, this variant has been classified as Pathogenic. This sequence change inserts 2 nucleotides in exon 17 of the RB1 mRNA (c.1673_1674dupTG), causing a frameshift at codon 559. This creates a premature translational stop signal (p.Glu559Trpfs*53) and is expected to result in an absent or disrupted protein product.
