This variant lies in the RB1 gene (transcript NM_000321.3) at coding-DNA position 967, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 323 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Glu323*) in the RB1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RB1 are known to be pathogenic (PMID: 17096365). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individuals with retinoblastoma (PMID: 12541220, 24791139, 25754945, 26396485). Invitae Evidence Modeling of clinical and family history, age, sex, and reported ancestry of multiple individuals with this RB1 variant has been performed. This variant is expected to be pathogenic with a positive predictive value of at least 99%.
