Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.6514G>C (p.Val2172Leu), citing Ambry Variant Classification Scheme 2023: The p.V2172L variant (also known as c.6514G>C), located in coding exon 39 of the DNAH11 gene, results from a G to C substitution at nucleotide position 6514. The valine at codon 2172 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
