Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_020297.4(ABCC9):c.215G>A (p.Arg72Lys), citing Ambry Variant Classification Scheme 2023: The p.R72K variant (also known as c.215G>A), located in coding exon 2 of the ABCC9 gene, results from a G to A substitution at nucleotide position 215. The arginine at codon 72 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
