Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020774.4(MIB1):c.2930G>A (p.Cys977Tyr), citing Ambry Variant Classification Scheme 2023: The p.C977Y variant (also known as c.2930G>A), located in coding exon 21 of the MIB1 gene, results from a G to A substitution at nucleotide position 2930. The cysteine at codon 977 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
