NM_152383.5(DIS3L2):c.305G>C (p.Arg102Pro) was classified as Uncertain significance for Perlman syndrome by Sema4, Sema4, citing Sema4 Curation Guidelines. This variant lies in the DIS3L2 gene (transcript NM_152383.5) at coding-DNA position 305, where G is replaced by C; at the protein level this means replaces arginine at residue 102 with proline — a missense variant. Submitter rationale: The DIS3L2 c.305G>C (p.R102P) variant has not been reported in the literature to our knowledge. It was observed in 7/10292 chromosomes of the Ashkenazi Jewish subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 410747). In silico tools suggest the impact of the variant on protein function is deleterious, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
