Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.620_621delinsT (p.Pro207fs), citing Ambry Variant Classification Scheme 2023: The c.620_621delCAinsT variant, located in coding exon 1 of the MET gene, results from the deletion of two nucleotides and insertion of one nucleotide causing a translational frameshift with a predicted alternate stop codon (p.P207Lfs*10). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. However, loss of function of MET has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
