Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_178452.6(DNAAF1):c.1033G>C (p.Glu345Gln), citing Ambry Variant Classification Scheme 2023. This variant lies in the DNAAF1 gene (transcript NM_178452.6) at coding-DNA position 1033, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 345 with glutamine — a missense variant. Submitter rationale: The p.E345Q variant (also known as c.1033G>C), located in coding exon 8 of the DNAAF1 gene, results from a G to C substitution at nucleotide position 1033. The glutamic acid at codon 345 is replaced by glutamine, an amino acid with highly similar properties. This alteration was reported in an individual with recurrent respiratory tract infection and severe early-onset obesity. The individual also had c.1698+1G>A in the DNAAF1 gene; however, the phase is unknown (Paz-Filho G et al. Genes (Basel), 2014 Aug;5:709-25). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
