Submitter rationale: The p.R45C variant (also known as c.133C>T), located in coding exon 1 of the BAG3 gene, results from a C to T substitution at nucleotide position 133. The arginine at codon 45 is replaced by cysteine, an amino acid with highly dissimilar properties. This alteration has been reported in a dilated cardiomyopathy (DCM) cohort; however, clinical details were limited (Mazzarotto F et al. Circulation, 2020 Feb;141:387-398).
