While this particular variant has not been reported in the literature, loss-of-function variants in PALB2 are known to be pathogenic (PMID:25099575, 17200668). This sequence change duplicates 1 nucleotide in exon 4 of the PALB2 mRNA (c.284dupA), causing a frameshift at codon 96. This creates a premature translational stop signal (p.Thr96Aspfs*6) and is expected to result in an absent or disrupted protein product.
