NM_007194.4(CHEK2):c.586A>G (p.Asn196Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N196D variant (also known as c.586A>G), located in coding exon 3 of the CHEK2 gene, results from an A to G substitution at nucleotide position 586. The asparagine at codon 196 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
