Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_014391.3(ANKRD1):c.211A>G (p.Lys71Glu), citing Ambry Variant Classification Scheme 2023: The p.K71E variant (also known as c.211A>G), located in coding exon 3 of the ANKRD1 gene, results from an A to G substitution at nucleotide position 211. The lysine at codon 71 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
