Pathogenic for Flattened knee epiphyses; Flattened femoral head; Genu valgum; Knee contracture; Metacarpophalangeal joint contracture; Osteoarthritis, hip; Multiple epiphyseal dysplasia type 4 — the classification assigned by Laboratory of Functional Genomics, Research Centre for Medical Genetics to NM_000112.4(SLC26A2):c.398C>T (p.Ala133Val), citing ACMG Guidelines, 2015: This variant is present in the gnomAD v4.1.0 database with a total AF of 2.48e-6 (4/1613306 alleles). The pathogenic effect of the variant is confirmed by the results of more than three in silico prediction programs (MetaLR 0.91). It was identified in a compound heterozygous state with p.Cys653Ser in one patient with rMED. Furthermore, this variant was reported by Panzer et al., 2008 in a patient with a p.Arg178Ter variant, presenting an intermediate phenotype between Desbuquois Dysplasia and diastrophic dysplasia (DTD).
