Uncertain significance for Hypertrophic cardiomyopathy 13; Dilated cardiomyopathy 1Z — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003280.3(TNNC1):c.207C>G (p.Ser69Arg), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TNNC1 gene (transcript NM_003280.3) at coding-DNA position 207, where C is replaced by G; at the protein level this means replaces serine at residue 69 with arginine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a TNNC1-related disease. In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. This sequence change replaces serine with arginine at codon 69 of the TNNC1 protein (p.Ser69Arg). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and arginine.
