Uncertain significance for Hypertrophic cardiomyopathy 13; Dilated cardiomyopathy 1Z — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003280.3(TNNC1):c.175dup (p.Glu59fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TNNC1 gene (transcript NM_003280.3) at coding-DNA position 175, duplicating one base; at the protein level this means shifts the reading frame starting at glutamic acid residue 59, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Glu59Glyfs*5) in the TNNC1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in TNNC1 cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TNNC1-related conditions. ClinVar contains an entry for this variant (Variation ID: 409872). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
