NM_000059.4(BRCA2):c.7958T>G (p.Leu2653Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L2653R variant (also known as c.7958T>G), located in coding exon 16 of the BRCA2 gene, results from a T to G substitution at nucleotide position 7958. The leucine at codon 2653 is replaced by arginine, an amino acid with dissimilar properties. The results from two saturation genome editing-based studies, including a haploid cell-survival assay and a humanized mouse embryonic stem cell line assay of drug response and survival, are discordant for this nucleotide substitution (Huang H et al. Nature. 2025 Feb;638(8050):528-537; Sahu S et al. Nature. 2025 Feb;638(8050):538-545). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
