NM_001098511.3(KIF2A):c.824G>C (p.Arg275Pro) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.824G>C (p.R275P) alteration is located in exon 9 (coding exon 9) of the KIF2A gene. This alteration results from a G to C substitution at nucleotide position 824, causing the arginine (R) at amino acid position 275 to be replaced by a proline (P). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
