Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.1585A>G (p.Lys529Glu), citing Ambry Variant Classification Scheme 2023: The p.K529E variant (also known as c.1585A>G), located in coding exon 11 of the PTCH1 gene, results from an A to G substitution at nucleotide position 1585. The lysine at codon 529 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
