In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a COL5A1-related disease. This sequence change replaces glutamic acid with lysine at codon 1499 of the COL5A1 protein (p.Glu1499Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine.
