Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000238.4(KCNH2):c.2649_2654dup (p.Arg885_Lys886insGlnArg), citing Ambry Variant Classification Scheme 2023. This variant lies in the KCNH2 gene (transcript NM_000238.4) at coding-DNA position 2649 through coding-DNA position 2654, duplicating 6 bases. Submitter rationale: The c.2649_2654dupGCAACG variant (also known as p.Q884_R885dup), located in coding exon 11 of the KCNH2 gene, results from an in-frame duplication of GCAACG at nucleotide positions 2649 to 2654. This results in the duplication of 2 extra residues (QR) between codons 884 and 885. This amino acid region ranges from highly conserved to not well conserved in available vertebrate species. In addition, the in silico prediction for this variant is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
