Uncertain significance for Oligodontia-cancer predisposition syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004655.4(AXIN2):c.1384C>A (p.Pro462Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with threonine at codon 462 of the AXIN2 protein (p.Pro462Thr). The proline residue is highly conserved and there is a small physicochemical difference between proline and threonine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a AXIN2-related disease. In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies.
