Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.911G>A (p.Ser304Asn), citing Ambry Variant Classification Scheme 2023: The p.S304N variant (also known as c.911G>A), located in coding exon 2 of the AXIN2 gene, results from a G to A substitution at nucleotide position 911. The serine at codon 304 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
