This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 1213, where A is replaced by C; at the protein level this means replaces serine at residue 405 with arginine — a missense variant. Submitter rationale: GLA c.1213A>C is a missense variant that changes the amino acid at residue 405 from Serine to Arginine. To our knowledge, this variant has not been reported in patients affected with Fabry disease in the published literature. Functional studies have been reported; however, the significance of the findings remain unclear (PMID:32023956;27657681;23935525). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Ser405Arg (c.1213A>C) as a variant of unknown significance.
