Submitter rationale: GLA c.960T>G is a missense variant that changes the amino acid at residue 320 from Asparagine to Lysine. This variant has been observed in at least one proband affected with Fabry disease (PMID:30386727;9105656;7575533;19285316;9213168). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:27657681). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify GLA c.960T>G as a likely pathogenic variant.
