Likely pathogenic for Fabry disease — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000169.3(GLA):c.827G>C (p.Ser276Thr), citing Genomenon Sequence Variant Interpretation Standards: GLA c.827G>C is a missense variant that changes the amino acid at residue 276 from Serine to Threonine. This variant has been observed in at least one proband affected with Fabry disease (PMID:32843101). Functional studies have been reported; however, the significance of the findings remain unclear and/or were performed in patient cells (PMID:32843101). The presence of pathogenic/likely pathogenic missense variant(s) at the same amino acid position indicates that this residue is likely important for protein function. It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify GLA c.827G>C as a likely pathogenic variant.
