Uncertain significance for Fabry disease — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000169.3(GLA):c.471G>C (p.Gln157His), citing Genomenon Sequence Variant Interpretation Standards: GLA c.471G>C is a missense variant that changes the amino acid at residue 157 from Glutamine to Histidine. This variant has been observed in at least one proband affected with Fabry disease (PMID:32583479;35971858;30506669). Functional studies have been reported; however, the significance of the findings remain unclear and/or were performed in patient cells (PMID:30506669). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify GLA c.471G>C as a variant of unknown significance.
