This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 352, where C is replaced by A; at the protein level this means replaces arginine at residue 118 with serine — a missense variant. Submitter rationale: GLA c.352C>A is a missense variant that changes the amino acid at residue 118 from Arginine to Serine. To our knowledge, this variant has not been reported in patients affected with Fabry disease in the published literature. Functional studies have been reported; however, the significance of the findings remain unclear and/or were performed in patient cells (PMID:23935525). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA c.352C>A as a variant of unknown significance.
