Likely pathogenic for Fabry disease — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000169.3(GLA):c.334C>G (p.Arg112Gly), citing Genomenon Sequence Variant Interpretation Standards. This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 334, where C is replaced by G; at the protein level this means replaces arginine at residue 112 with glycine — a missense variant. Submitter rationale: GLA c.334C>G is a missense variant that changes the amino acid at residue 112 from Arginine to Glycine. This variant has been observed in at least one proband affected with Fabry disease (PMID:27657681;27834756). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:27657681). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify GLA c.334C>G as a likely pathogenic variant.
