This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 243, where G is replaced by T; at the protein level this means replaces tryptophan at residue 81 with cysteine — a missense variant. Submitter rationale: GLA p.Trp81Cys (c.243G>T) is a missense variant that changes the amino acid at residue 81 from Tryptophan to Cysteine. To our knowledge, this variant has not been reported in patients affected with Fabry disease in the published literature. At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:27657681). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify GLA p.Trp81Cys (c.243G>T) as a likely pathogenic variant.
