Pathogenic for Fabry disease — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000169.3(GLA):c.184dup (p.Ser62fs), citing Genomenon Sequence Variant Interpretation Standards: GLA p.Ser62PhefsTer5 (c.184dup) is a frameshift variant that results in the production of a truncated protein which is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID:32843101). The variant was found to segregate with disease in at least one affected family (PMID:32843101). Functional studies have been reported; however, the significance of the findings remain unclear and/or were performed in patient cells (PMID:32843101;33016649). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Ser62PhefsTer5 (c.184dup) as a pathogenic variant.
