This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 77, duplicating one base; at the protein level this means shifts the reading frame starting at glycine residue 28, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: GLA p.Gly28TrpfsTer3 (c.77dup) is a frameshift variant that results in the production of a truncated protein that may be subject to nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID: 32991443; 39609713). Functional studies have been reported; however, the significance of the findings remain unclear and/or were performed in patient cells (PMID: 32991443; 39609713). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Gly28TrpfsTer3 (c.77dup) as a pathogenic variant.
