This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 319, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 107 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: GLA p.Gln107Ter (c.319C>T) is a nonsense variant that introduces a premature stop codon at amino acid position 107, creating a truncated protein that is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID:29853467;23826564;25149322;16151903;34440358;20576773;29661900;39362930;23546814). Functional studies have been reported; however, the significance of the findings remain unclear and/or they were performed in patient cells (PMID:16151903). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Gln107Ter (c.319C>T) as a pathogenic variant.
