This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 264, where C is replaced by G; at the protein level this means converts the codon for tyrosine at residue 88 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: GLA p.Tyr88Ter (c.264C>G) is a nonsense variant that introduces a premature stop codon at amino acid position 88, creating a truncated protein that is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID:32843101). The variant was found to segregate with disease in at least one affected family (PMID:32843101). Functional studies have been reported; however, the significance of the findings remain unclear and/or they were performed in patient cells (PMID:32843101). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Tyr88Ter (c.264C>G) as a pathogenic variant.
