This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 168, where C is replaced by A; at the protein level this means converts the codon for cysteine at residue 56 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: GLA p.Cys56Ter (c.168C>A) is a nonsense variant that introduces a premature stop codon at amino acid position 56, creating a truncated protein that is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID:16595074;33915609). Functional studies have been reported; however, the significance of the findings remain unclear and/or they were performed in patient cells (PMID:16595074;33915609). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Cys56Ter (c.168C>A) as a pathogenic variant.
